Simultaneous Pharmacokinetic and Pharmacodynamic Analysis of 5α-Reductase Inhibitors and Androgens by Liquid Chromatography Tandem Mass Spectrometry

    The study developed a novel assay using liquid chromatography tandem mass spectrometry (LC-MS/MS) to measure the concentrations of 5α-reductase inhibitors (finasteride and dutasteride), their substrates, and the product (DHT) in human serum. Tested on 16 male subjects aged 20-85 years, the method demonstrated precision, accuracy, and stability, requiring only 500 µL of serum. Both finasteride and dutasteride significantly suppressed DHT concentrations by approximately 47% while increasing testosterone levels. The assay provided a robust marker of treatment adherence and effectiveness, although further technological advances were suggested to enhance its sensitivity and measurable pharmacodynamic range.
