Establishment and Characterization of Matched Immortalized Human Frontal and Occipital Scalp Dermal Papilla Cell Lines from Androgenetic Alopecia

    The study established immortalized human frontal and occipital scalp dermal papilla (DP) cell lines from androgenetic alopecia (AGA) patients using the simian virus 40 large T antigen (SV40T-Ag) and human telomerase reverse transcriptase (hTERT). The frontal DP cells were found to be more sensitive to dihydrotestosterone (DHT), leading to hair follicle miniaturization and hair loss in AGA. They also expressed higher levels of androgen receptor (AR) and 5-alpha reductase type 2 (5αR2) compared to occipital DP cells. The frontal DP cells showed a slower proliferation rate, reduced expression of hair growth factors, and improper sphere formation due to increased expression of certain enzymes. These cell lines could provide insights into AGA pathogenesis and potentially lead to targeted therapies for hair loss.
