Engineered Exosomes Co-Delivering EGF and FGF Ameliorate Androgenetic Alopecia in a Mouse Model

    The study explores the use of engineered exosomes co-delivering Epidermal Growth Factor (EGF) and Fibroblast Growth Factor (FGF) to treat androgenetic alopecia (AGA) in a mouse model. The engineered exosomes, utilizing LAMP2B fusion technology, showed enhanced delivery and stability of EGF and FGF, promoting hair follicle regeneration. In vitro and in vivo experiments demonstrated increased proliferation and migration of human dermal papilla cells and improved hair density and coverage in AGA mice. Histological analysis confirmed an increased number of anagen-phase hair follicles. The treatment increased hair coverage from 21.5% to 58.4% and showed no toxicological damage to major organs, indicating its safety. These findings suggest that EXO-EGF/FGF could be a promising therapeutic approach for AGA by reversing androgen-driven suppression of hair growth factors.
