This document describes a technique for producing 3D spheroids of high-passaged human dermal papilla cells using surface tension guided hanging-drop. The spheroids can be used to form inductive microtissues for hair-follicle regeneration. The study found that the spheroids exhibited greater expression of signature genes and proteins associated with hair-inductivity compared to 2D cultured cells. The DP microtissues induced hair follicle neogenesis in vivo when co-transplanted with epidermal cells in nude mice. The study suggests that DP microtissues could be a promising approach for hair-follicle regeneration.
