Multi-View Light Sheet Fluorescence Microscopy for Imaging Cellular Self-Assembly in Spheroids of Human Hair Follicle Dermal Papilla Cells and Keratinocytes

    The study demonstrated that multi-view light sheet fluorescence microscopy (LSFM) is an effective tool for observing the self-assembly of human hair follicle dermal papilla cells and keratinocytes into spheroids. The cells, labeled with fluorescent dyes, were incubated to form spheroids and imaged using LSFM. Results showed that rat tail collagen 1 led to the formation of a single spheroid, while its absence resulted in multiple spheroids. After 2 days of incubation, dermal papilla cells were found in the core and keratinocytes on the periphery of the spheroids. This technique provided detailed 3D quantification of cell distribution, offering insights into the cellular dynamics of skin regeneration and hair follicle formation.
