The study demonstrates that Alpinetin (AP) promotes hair regeneration by activating hair follicle stem cells (HFSCs) and extending the anagen phase of the hair cycle in a mouse model. AP-treated mice showed increased cell proliferation in hair follicles, delayed entry into the catagen phase, and activation of Lgr5+ and Gli1+ HFSCs through the Wnt signaling pathway. RNA-seq analysis revealed significant upregulation of genes involved in hair cycle regulation and Wnt signaling. Importantly, AP exhibited no cytotoxicity in human and mouse skin cells, suggesting its potential as a safe and effective treatment for alopecia.
