The study investigates the dynamic interaction between IL-1 and WNT pathways in dermal adipocyte lineage cells during skin development and wound healing. Using single-cell RNA sequencing on mouse skin, researchers identified distinct cell states and found that IL-1-NF-κB signaling promotes adipogenesis, while WNT-β-catenin signaling inhibits it. Neutrophils mediate wound-induced adipogenesis through the IL-1R-NF-κB-CREB axis. Conversely, WNT activation inhibits adipogenesis and promotes lipolysis and dedifferentiation, leading to myofibroblast formation. This sustained WNT activation is also observed in human keloids.
