Follicular-Targeted Delivery of Spironolactone Provided by Polymeric Nanoparticles

    Ricardo Ferreira‐Nunes, Marcílio Cunha-Filho, Guilherme M. Gelfuso
TLDR Nanoparticles can effectively deliver spironolactone to hair follicles for treating alopecia and acne.
This study developed polymeric nanoparticles of different sizes to deliver spironolactone topically for treating acne and alopecia. The nanoparticles, made from poly-ε-caprolactone (PCL) or methacrylic acid/methyl methacrylate copolymer (EL100), were found to be chemically compatible with spironolactone, physically stable, and non-irritating. They effectively stabilized the drug for at least 90 days and controlled its release in aqueous media. Notably, larger nanoparticles (180 nm) significantly enhanced drug delivery to hair follicles compared to smaller ones, making them a promising option for targeted topical therapy.

Crushing spironolactone pills and combining them with minoxidil for topical application may be effective for hair loss, with the addition of finasteride suggested. Adding rosemary and peppermint oils can help mask the unpleasant smell.

Spironolactone can remain effective for a long time in treating androgenetic alopecia, but it may not completely stop hair loss. Hair loss treatments like spironolactone, finasteride, and dutasteride slow down hair loss rather than cure it, and their effectiveness can vary based on individual response and the aggressiveness of the condition.

Spironolactone is more potent and lowers testosterone and DHT, while dutasteride only impacts DHT. For female hair loss, checking hormones and considering treatments like oral minoxidil, spironolactone, or topical minoxidil is recommended.
