TLDR TLR2 is important for hair growth and can be targeted to treat hair loss.
Our study reveals that TLR2 plays a crucial role in maintaining hair follicle homeostasis and regeneration by regulating HFSCs through BMP signaling. TLR2 expression is cycle-dependent and essential for HFSC activation. Aging and obesity reduce TLR2 and its ligand CEP, impairing hair regeneration. Administering CEP can stimulate hair growth via TLR2, highlighting a new link between innate immunity and hair follicle health, offering potential new treatments for hair loss.
