Nanostructured Lipid Carriers Promote Percutaneous Absorption and Hair Follicle Targeting of Tofacitinib for Treating Alopecia Areata

    The study developed tofacitinib-loaded cationic lipid nanoparticles (TFB-cNLCs) to enhance percutaneous absorption and hair follicle targeting for treating alopecia areata. Using ex vivo pig ear and in vitro follicle models, TFB-cNLCs demonstrated improved drug delivery to hair follicles, reduced IFN-γ-induced symptoms, and decreased CD8+NKG2D+T cells in a C3H mouse model, effectively reversing hair loss. This formulation offers a promising topical treatment, minimizing systemic side effects while maintaining efficacy.
