The study introduces a novel method using Ficoll density gradient sedimentation (FDGS) to isolate pelage hair follicle mesenchymal stem cells (HF-MSCs) from adult mouse back skin. This method is more efficient than traditional techniques, yielding a larger number of cells with fewer mice. The isolated HF-MSCs exhibited similar markers, growth patterns, and hair follicle induction abilities as whisker HF-MSCs. When injected into mice, these cells promoted hair growth by secreting exosomes, which likely target the dermal papilla and hair matrix regions. This method offers a quick and convenient way to obtain HF-MSCs for experimental research and potential therapeutic applications.
