The Bioengineering of Microspheric Skin Organoids and Their Application in Drug Screening

    This study presents a novel method for creating microspheric skin organoids using spinning bioreactors, which mimic the bilayered structure of human skin. The organoids consist of a core-shell structure with human keratinocytes forming the epidermis and a core of human dermal fibroblasts and collagen representing the dermis. These organoids are designed for high-throughput drug screening and are equipped with a luciferase reporter to detect Wnt pathway activation. The study identifies Minoxidil as a compound that activates the epidermal Wnt/beta-catenin pathway, demonstrating the organoids' potential for drug testing applications.
