The study "TLR2 Regulates Hair Follicle Cycle and Regeneration via BMP Signaling" reveals that the maintenance and regeneration of hair follicles depend on TLR2 (Toll-like receptor 2) in hair follicle stem cells (HFSCs). TLR2 is expressed in a cycle-dependent manner in healthy hair follicles and regulates HFSC activation by counteracting inhibitory BMP signaling. Aging and obesity are associated with a decrease in both TLR2 and its endogenous ligand carboxyethylpyrrole (CEP), a metabolite of polyunsaturated fatty acids, which may lead to diminished hair growth. However, administering CEP can stimulate hair regeneration through a TLR2-dependent mechanism. This research establishes a new link between TLR2-mediated innate immunity and HFSC activation, which is crucial for hair follicle health and preventing hair loss.
