The study aimed to optimize the culture conditions for goat hair follicle stem cells (gHFSCs) to facilitate research on hair growth and biology. The researchers tested the effects of the Rho-associated protein kinase inhibitor Y-27632, leukemia inhibitory factor (LIF), basic fibroblast growth factor (bFGF), and vitamin C on gHFSC proliferation and pluripotency. They found that Y-27632, LIF, and bFGF each independently improved gHFSC proliferation and pluripotency, with Y-27632 having the most significant effect. The combination of Y-27632 (10 μM) and bFGF (10 ng/mL) provided the best results for cell proliferation, clone-forming ability, and differentiation potential. This optimized culture condition, named goat Hair Follicle Enhanced Medium (gHFEM), is considered ideal for in vitro gHFSC culture, which is crucial for studying hair follicle growth and function.
