This study investigated the correlation between polymorphisms in four keratin-associated protein (KRTAP) genes and cashmere fiber diameters in Jiangnan and Tibetan cashmere goats. Using multiplex PCR and exome sequencing, 28 mutation sites were identified across the KRTAP15-1, KRTAP13.1, KRTAP27-1, and KRTAP24-1 genes. Significant correlations were found between the polymorphisms in KRTAP13.1, KRTAP27-1, and KRTAP24-1 and the fiber diameter in Jiangnan cashmere goats. The findings suggest that these gene polymorphisms can serve as molecular markers to improve cashmere fiber diameter, aiding in the breeding and conservation of high-quality cashmere goats. The study involved 353 Jiangnan and 299 Tibetan cashmere goats.
