Generating Hair Follicle Inductive Dermal Papillae Cells From Adipose-Derived Mesenchymal Stem Cells

This study aimed to achieve de novo folliculogenesis by generating dermal papillae (DP)-like spheroids from adipose-derived mesenchymal stem cells (ASCs). The researchers used the hanging drop method and Wnt exposure to direct ASCs towards a DP-like fate, mimicking embryological hair follicle induction. Gene expression analysis showed a significant increase in the DP-cell marker Versican when ASCs were exposed to Wnt, suggesting that Wnt/β-catenin signaling regulates DP cell growth. The resulting spheroids exhibited similar morphology and gene expression to human hair follicle DP cells. This method of generating DP-like cells from readily obtainable ASCs could potentially be rapidly translatable to clinical trials for hair follicle formation.
