The Myeloid Mineralocorticoid Receptor Regulates Dermal Angiogenesis and Inflammation in Glucocorticoid-Induced Impaired Wound Healing

    The study investigated the effects of mineralocorticoid receptor (MR) blockade on wound healing in C57Bl6 mice pretreated with the dermocorticoid clobetasol. It was found that topical MR inhibition using canrenoate improved delayed wound healing by resolving prolonged inflammation and promoting angiogenesis. This was achieved by increasing the ratio of anti-inflammatory to pro-inflammatory macrophages and upregulating pro-angiogenic factors. Additionally, genetic deletion of MR in myeloid cells mirrored the benefits of pharmacological MR blockade, suggesting that MR antagonism facilitates macrophage phenotype switching to enhance wound healing impaired by glucocorticoid treatment.
