TLDR Keratin 75 is important for fast wound healing and works with SOX2 and the LINC complex to help skin cells move and repair damage.
The document presents a study on the role of Keratin 75 (K75) in wound healing, revealing that K75 is a direct target of the transcription factor SOX2, which is known for promoting rapid wound repair in the oral epithelium. The study utilized a tamoxifen-inducible mouse model to show that SOX2 upregulates K75 expression in the interfollicular epidermis. Additionally, K75 was found to be a part of the linker of nucleoskeleton and cytoskeleton (LINC) complex, which is responsible for transmitting mechanical signals that influence gene expression. The interaction between K75 and SUN2, a LINC complex component, was shown to be crucial for the SOX2-mediated rapid healing response. Knockdown experiments of K75 or SUN2 disrupted keratinocyte migration and negated the healing effects of SOX2 overexpression, underscoring the importance of K75 in wound healing beyond its structural role in hair, teeth, and feathers.
