The study found that Necrostatin-1s (Nec-1s), a RIPK1 inhibitor, effectively promoted hair growth by enhancing the proliferation and migration of human outer-root sheath (ORS) cells and increasing hair follicle length in mouse and pig organ cultures. Nec-1s accelerated the transition from the telogen to anagen phase and extended the anagen period in mice. It activated the Wnt/β-catenin signaling pathway, crucial for hair growth, by upregulating Wnt3a and Wnt5b mRNA expression. The study suggested that Nec-1s could be a potential treatment for hair loss by inhibiting necroptosis and activating hair growth pathways, as RIPK1 and RIPK3 were highly expressed during hair regression.
