The study investigates the effects of BM15766 and 7DHC on hair follicle organoid cultures, focusing on the hair follicle bulge, a key stem cell region. Both compounds caused structural damage, disrupted cellular organization, and led to the deterioration of the hair matrix, pushing the hair fiber upwards. Real-time PCR showed significant down-regulation of SOX9 and LGR5 gene expression in treated samples compared to controls. Immunofluorescence imaging revealed reduced LGR5 and K15 expression and increased apoptosis in treated hair follicles. TEM analysis showed that treated samples had ruptured matrix cells, fragmented keratin filaments, and degraded connective tissue sheaths, indicating significant structural damage compared to well-organized control samples.
