Determination of Vinflunine in Beagle Dog's Plasma by LC-MS and Study of Its Pharmacokinetics

The study developed a method using LC-MS to quantify vinflunine in Beagle dog plasma and examined its pharmacokinetics. The method involved spiking plasma with finasteride as an internal standard, alkalizing it, and extracting it with ethyl acetate. The analysis was performed on a Shimadzu VP-ODS column with a methanol-ammonium acetate buffer mobile phase. The method showed linear calibration plots for vinflunine concentrations ranging from 0.025 to 12.5 μg/mL, with intra-assay and inter-assay variability under 7.8% and a relative recovery rate between 97.0% and 100.2%. The study concluded that the LC-MS method was simple, accurate, sensitive, and suitable for pharmacokinetic studies of vinflunine in dogs.
