TLDR Sorafenib causes skin reactions by increasing the number and activity of skin mast cells.
The study investigated the effects of sorafenib on skin-type mast cells (MCs) and found that it significantly increased the number and degranulation of MCs in human skin samples, without affecting their proliferation and apoptosis. This indicated that sorafenib stimulated MC maturation from resident precursors through PI3K signaling and increased stem cell factor (SCF) expression. The findings suggested that targeting PI3K signaling or SCF could be potential strategies for managing sorafenib-induced skin reactions.
