TLDR Ruxolitinib helps protect skin stem cells and keeps skin healthy in mice with skin GVHD.
The study demonstrated that topical ruxolitinib protected Lgr5+ hair follicle stem cells (HFSCs) and maintained skin homeostasis in murine graft-versus-host disease (GVHD). Unlike corticosteroids, which caused HFSC loss and adverse skin effects, ruxolitinib suppressed donor T cell infiltration and inflammation without damaging skin stem cells. This suggested that ruxolitinib could be a superior treatment for inflammatory skin diseases, including GVHD and alopecia areata, by promoting hair growth and avoiding the complications associated with systemic administration. The findings highlighted a novel mechanism of skin injury in GVHD and proposed a promising therapeutic strategy using JAK inhibitors.

Key treatments for hair preservation include minoxidil, finasteride/dutasteride, and microneedling, with additional options like specific shampoos, oils, biotin, zinc, tretinoin, clascoterone, and low-level laser therapy. Maintaining a healthy lifestyle, reducing stress, and addressing nutrient deficiencies can also support hair health.
