An Investigation of Keratin 15 Function by Small Interfering RNA Technology

    The study investigated the role of keratin 15 (K15) in maintaining the stem cell niche by using siRNA to knock down K15 in various cell lines. It was found that K15 knock-down did not affect cell adhesion but did influence cell spreading, morphology, migration, differentiation, and proliferation. Specifically, K15-deficient cells spread more quickly, were larger, less motile, showed increased differentiation as indicated by higher involucrin expression, and proliferated more than control cells. K15 was present in the basal layers of the epidermis and around hair follicles in rat, mouse, and human skin. The study concluded that K15 impacts the stem cell characteristics of keratinocytes, though further research was needed to understand the underlying mechanisms.
