The study aimed to determine the best culture conditions for growing epithelial stem cells (ESCs) from plucked hair follicles. It compared various conditions, including no coating, rat tail-collagen coating, collagen Ⅳ coating, and 3T3 fibroblasts feeder. The results showed that the optimal condition for ESC growth was using the 3T3 fibroblasts feeder, particularly for cells from the bulge area of the hair follicle. This was confirmed by RT-PCR and immunofluorescent staining, which detected CK15, a marker specific to ESCs.
