Comparative Study on Isolation and Culture Methods of Hair Follicle Stem Cells in Fine-Wool Sheep

    The study focused on isolating and culturing hair follicle stem cells from fine-wool sheep to establish an in vitro culturing system. Three methods were tested: two-step enzyme digestion, mechanical separation, and a combination of both. The cells were evaluated based on number, cloning efficiency, and proliferation ability. All methods successfully produced cells expressing keratin K19 and integrin-β1, confirming the successful isolation of hair follicle stem cells. The combination of two-step enzymatic digestion and mechanical separation was found to be the optimal method for obtaining and culturing these cells.
