Characterization of a Cluster of Human High/Ultrahigh Sulfur Keratin-Associated Protein Genes Embedded in the Type I Keratin Gene Domain on Chromosome 17q12-21

    TLDR A cluster of sulfur-rich hair protein genes was found on chromosome 17.
    Researchers used a human hair keratin-associated protein (hKAP1.1A) gene probe to screen a human P1 artificial chromosome library, isolating six P1 clones. These clones were found within four previously sequenced human bacterial artificial chromosome clones on chromosome 17q12-21, forming two large contigs of 290 and 225 kilobase pairs (kb). A fifth clone sequence closed these contigs into a 600-kb cluster, which included six type I hair keratin genes at one end and type I cytokeratin K20 and K12 genes at the other. The cluster's center contained 37 high/ultrahigh sulfur hair keratin-associated protein (KAP) genes, divided into seven multigene families. Screening a human scalp cDNA library identified 26 human KAP cDNA clones and four additional cDNA sequences from KAP gene families. In situ hybridization localized mRNAs of five KAP multigene families to the upper cortex of the hair shaft.
