The study explored how Bone Morphogenetic Protein-2 (BMP-2) regulated the Dlx3 gene in mouse keratinocytes and hair follicle cells. BMP-2 was shown to induce Dlx3 transcription within 12 hours by binding Smad1 and Smad4 to a specific GCAT sequence in the Dlx3 promoter. This mechanism was independent of the keratinocytes' differentiation state and provided an alternative to the known PKC-dependent Ca2+ activation pathway. The findings emphasized the role of BMP signaling in Dlx3 expression, crucial for hair follicle development and epidermal differentiation.
