TLDR Tissue stem cells originate from specific areas in organs and are vital for organ maintenance and repair.
This review examined the developmental origins of tissue stem cells in various organs, focusing on hair follicles, intestines, brain, skeletal muscles, and the hematopoietic system. It highlighted the role of genetic lineage tracing and single-cell transcriptomics in understanding stem cell dynamics. Specifically, for hair follicle stem cells, the study identified Sox9-positive cells as precursors originating from a peripheral ring zone, challenging previous beliefs. The "telescope model" was introduced to describe hair follicle development, emphasizing the importance of tissue architecture in stem cell fate. The review underscored the significance of integrating molecular, cellular, and mechanical systems to understand organ-specific morphogenetic processes and suggested that emerging technologies could further elucidate the coordination between tissue patterning and stem cell development.
