Therapeutic deep eutectic solvents (TheDES) are highlighted as a promising solution for enhancing the solubility and transdermal delivery of active pharmaceutical ingredients (APIs), addressing the challenge posed by the stratum corneum barrier. The review explores the physicochemical properties of TheDES, their permeation enhancement mechanisms, including the "lipid exchange" effect, and their evolution into advanced forms like eutectogels and nanoemulsions. TheDES show potential in treating pain, infectious diseases, and chronic conditions, with considerations for dermatological safety and future computational modeling developments.
