TLDR Girard's reagent P improves detection of spironolactone and its metabolites.
This study investigates the use of Girard's reagent P (GP) to derivatize spironolactone and its metabolites, enhancing their detection and analysis using mass spectrometry and ion mobility spectrometry. Spironolactone, used for conditions like alopecia, is difficult to analyze due to rapid metabolism and similar metabolites. GP derivatization improves sensitivity and selectivity by reducing in-source fragmentation and enhancing ionization efficiency. The study successfully characterizes these compounds using advanced spectrometry techniques and validates the method in murine serum, demonstrating its potential for pharmacokinetic studies. The research highlights the need for further optimization and development of quantitative methods.
