TLDR The method and source of keratinocytes affect the structure of reconstructed skin.
The study investigated the architecture of reconstructed epidermis on collagen lattices, revealing that the method and source of keratinocytes significantly affected the results. Epidermis was cultured from either interfollicular epidermis or hair follicle outer root sheath, and differences were noted in the expression of 67 kDa keratin. Notably, only epidermis derived from hair follicle explants showed a suprabasal and homogeneous expression of this keratin, similar to normal epidermis. Other methods resulted in delayed and patchy expression. These findings suggested intrinsic differences between interfollicular and follicular keratinocytes.
