Isoxazole 9 (ISX9), a small molecule targeting Axin, activates Wnt/β‐catenin signalling and promotes hair regrowth

    The study identified Isoxazole 9 (ISX9) as a novel activator of the Wnt/β-catenin signaling pathway, crucial for hair growth and regeneration. ISX9 targeted Axin1, enhancing the LRP6-Axin1 association and stabilizing β-catenin, which promoted hair follicle proliferation and hair growth in C57BL/6 mice. The study found that ISX9 was more effective than minoxidil in promoting hair regrowth, with no significant adverse effects on major organs. ISX9 upregulated Wnt target genes and stemness marker genes, suggesting its potential as a therapeutic agent for alopecia treatment. Further studies were needed to evaluate its efficacy and safety in humans.
