Iontophoretic Transdermal Delivery Using Chitosan-Coated PLGA Nanoparticles for Positively Charged Drugs

    TLDR Chitosan-coated nanoparticles can effectively deliver positively charged drugs through the skin using iontophoresis.
    The study prepared Donepezil hydrochloride (DP)-loaded PLGA nanoparticles coated with chitosan (CS) to achieve a positive surface charge, enhancing their suitability for iontophoretic transdermal delivery. The CS-coated nanoparticles had an average diameter of 117.7 ± 60.6 nm and showed a 2.2-fold increase in DP skin accumulation compared to non-coated nanoparticles when iontophoresis was applied. This indicated that the chitosan coating improved the nanoparticles' ability to penetrate the skin and deliver positively charged hydrophilic drugs effectively, reaching deep hair follicles and the stratum corneum.
