Gene Expression Profiling of Human Keratinocytes Exposed to the Hair-Dyeing Agent, m-Aminophenol

    The study investigated the effects of m-aminophenol (MAP), a common hair dye ingredient, on human keratinocytes (HaCaT cells) by analyzing changes in gene expression. Exposure to 1.2 mM MAP resulted in the up-regulation of 3,441 genes and down-regulation of 2,583 genes, with significant involvement in inflammatory and oxidative stress responses, as well as the TGF-beta signaling pathway. MAP exposure also induced reactive oxygen species (ROS) generation, contributing to cellular toxicity. The findings suggested that MAP-induced gene expression changes could serve as biomarkers for detecting skin toxicity, highlighting the potential risks associated with MAP in hair dyes.
