    This study utilized single-cell RNA sequencing and ATAC sequencing to analyze peripheral blood mononuclear cells from 6 mild alopecia areata (AA) patients, 5 severe AA patients, and 5 healthy controls, identifying 16 unique cell clusters among 32,084 cells. The research highlighted that over 80% of these cell types shared open chromatin regions linked to nine key transcription factors involved in various immune functions, such as T cell activation, dendritic cell maturation, and B cell maturation. The findings provide a detailed view of the transcriptional and epigenetic differences between mild and severe AA, offering insights for future research and potential therapeutic targets.
