This study examined the association of androgen-related genes CYP21A2 and CYP19A1 with severe acne vulgaris in 1,252 Han Chinese individuals, including 600 patients and 652 controls. Significant associations were found between severe acne and SNPs in the CYP21A2 gene, particularly rs6474 and rs6465, with rs6474 linked to a higher risk and rs6465 having a protective effect. In males, SNPs in both CYP21A2 and CYP19A1 were associated with severe acne, with specific haplotypes influencing risk levels. No significant associations were found in females. The study suggested that genetic variations in these genes might affect androgen and estrogen levels, contributing to acne susceptibility, and highlighted the need for further research to understand these genetic mechanisms.
