This comprehensive review examines the role of transient receptor potential vanilloid (TRPV) channels in osteoarthritis (OA), focusing on their involvement in pain perception, inflammation, and mechanotransduction. TRPV channels, particularly TRPV1 and TRPV4, are highlighted as key players in OA pathogenesis, influencing pain generation and disease progression through mechanisms like cell necrosis and synovial inflammation. The review identifies TRPV channels as promising therapeutic targets, with studies showing that TRPV1 antagonists can effectively reduce pain. However, challenges such as adverse reactions and unclear molecular mechanisms remain. Future research should focus on developing selective TRPV modulators and exploring combination therapies to improve OA management.
