Role of STAT3 in Keratinocyte Stem Cells During Skin Tumorigenesis

The study investigated the role of Stat3 in keratinocyte stem cells (KSCs) during skin tumorigenesis. Deleting Stat3 in the bulge region of hair follicles in mice reduced tumor development by affecting KSC survival. In transgenic mice expressing active Stat3 (Stat3C), there was a decrease in CD34 and α6 integrin positive cells, with an increase in progenitor populations, suggesting altered cell migration and differentiation. Stat3C expression was linked to changes in β-catenin and α6-integrin, reducing cell adhesion and promoting migration. Additionally, Stat3 deletion in epidermal keratinocytes increased differentiation marker expression. The findings indicated that Stat3 influenced keratinocyte differentiation and migration, impacting epithelial carcinogenesis.
