Role of STAT3 in Keratinocyte Stem Cells During Skin Tumorigenesis

The study investigated the role of Stat3 in keratinocyte stem cells (KSCs) during skin tumorigenesis. It was found that deleting Stat3 in the bulge region of hair follicles reduced tumor development by affecting KSC survival. In transgenic mice expressing a constitutively active form of Stat3 (Stat3C), there was a decrease in stem cell markers (CD34 and α6 integrin) and an increase in progenitor populations, suggesting that Stat3C may induce migration of KSCs by altering cell adhesion. Additionally, Stat3 deletion in the epidermis led to increased differentiation of keratinocytes. These findings indicated that Stat3 played a significant role in keratinocyte differentiation and migration, which could influence epithelial carcinogenesis.
