Predictive Skin Permeation of Ethinylestradiol, α-Asarone, Finasteride, and Risperidone

    The study aimed to predict drug skin permeability using QSPR models and verify their accuracy for skin percutaneous penetration. Four drugs—ethinylestradiol, α-asarone, finasteride, and risperidone—were tested using seven QSPR models. The study combined drug physical-chemical parameters with these models to calculate skin penetration data, which were then compared to experimental results from in vitro penetration experiments using modified Franz-Diffusion Cells. The results showed that the log Jmax for the four drugs aligned with the 95% confidence interval predicted by the models. Models that used descriptors of hydrophobicity and molecular weight provided better predictions. The Potts-Guy model demonstrated the best prediction ability among the models tested.
