The study addresses the challenge of treating atopic dermatitis (AD) by developing a dissolvable microneedle (MN) patch that incorporates Portulaca oleracea L.-derived exosome-like nanoparticles (PELNs) and 4-amino-TEMPO conjugated with hyaluronic acid (HA-TEMPO). This combination aims to overcome the limited antioxidant activity and poor skin penetration of PELNs. The HA-TEMPO@PELN-MN system demonstrated effective reduction of cutaneous oxidative stress and suppression of pathological skin crusting in vivo, while also restoring Th1/Th2 immune balance. The MN patch offers a promising, biocompatible, and controlled strategy for AD therapy, with rapid drug release and efficient delivery.
