RGDSP-Functionalized Peptide Hydrogel Stimulates Growth Factor Secretion Via Integrin αv/PI3K/AKT Axis for Improved Wound Healing by Human Amniotic Mesenchymal Stem Cells

    This study examines the use of RGDSP-functionalized peptide hydrogels to enhance wound healing by human amniotic mesenchymal stem cells (hAMSCs). The hydrogels improve the secretion of growth factors like EGF, bFGF, VEGF, and TGF-β via the integrin αv/PI3K/AKT pathway, promoting wound re-epithelialization, angiogenesis, and epidermal maturation, including hair follicle formation. The study, conducted on a murine model, shows that the RGDmix hydrogel significantly accelerates wound healing, achieving complete wound closure by day 21. This research underscores the potential of these hydrogels in regenerative medicine by enhancing the paracrine function of hAMSCs and suggests further optimization for clinical applications.
