    This study presents a novel triple drug delivery system using 5-fluorouracil, curcumin, and piperine co-loaded human serum albumin nanoparticles (5FU-CUR-PIP-HSA-NPs) for breast tumor suppression. The system was developed using a scaffold-free platform to create homogenous tumor spheroids in standard 96-well plates, enhancing drug screening efficiency. The nanoparticles showed high drug loading efficiency (10%) and demonstrated effective cellular uptake and cytotoxicity in 3D tumor spheroids, highlighting their potential in anti-metastatic therapy. The study underscores the importance of 3D tumor models for preclinical drug screening, bridging the gap between in vitro and in vivo testing.
