The study explored the feasibility of quantitatively analyzing androgen metabolism by using single dermal papillae (DP) from human hair follicles, particularly in the context of androgenetic alopecia (AGA). AGA is characterized by the miniaturization of androgen-sensitive hair follicles, with increased 5α-reductase (5aR) activity being a key factor. The researchers employed a novel method using freshly isolated DP and a highly sensitive HPLC-radiomatic flow scintillation system to measure 5aR activity. They found that DP from the occipital scalp exhibited significant 5aR activity, although there was considerable variability in enzyme activity among individual DP. The study concluded that using only one or two DP is not currently a reliable method for analyzing 5aR activity ex vivo due to this variability.
