TLDR Increased Wnt signaling causes hair graying by depleting melanocyte stem cells in aged mice.
The study investigated the process of hair graying, or canities, in mice, focusing on the role of melanocytes and β-catenin expression during hair cycling. It was found that β-catenin expression increased in aged mice during both anagen and telogen phases compared to young mice. Overexpression of Wnt10b accelerated the anagen phase and promoted melanocyte differentiation in young mice, with increased β-catenin expression in specific hair follicle regions. This overexpression also enhanced melanocyte progenitor cell differentiation in vitro. The findings suggested that increased Wnt signaling led to excessive melanocyte differentiation, depleting melanocyte stem cells and resulting in hair graying in aged mice.
