Mouse Models of Alopecia Areata: C3H/HeJ Mice Versus the Humanized AA Mouse Model

    The document compared two mouse models for studying alopecia areata (AA): the C3H/HeJ mice and the humanized AA mouse model. The C3H/HeJ model, which develops AA-like symptoms spontaneously or through induction, has been instrumental in understanding AA's pathobiology and the role of psychoemotional stressors and IFN-γ. However, it was not ideal for testing certain immunoinhibitory therapies due to its histological differences from human AA. In contrast, the humanized AA model, which involves xenotransplanting human scalp onto SCID mice and inducing AA with IL-2-stimulated PBMCs, better mimicked human AA. It highlighted the importance of CD8+ T cells, NKG2D+ cells, and unconventional T cell subtypes in AA pathogenesis. This model was recommended for preclinical testing of new AA therapies due to its closer resemblance to human AA.
