TLDR A reliable method was developed to measure aristolochic acid-I in rat blood.
The study developed and validated a sensitive and specific liquid chromatography–tandem mass spectrometry method for determining aristolochic acid‐I (AA‐I) in rat plasma, using finasteride as an internal standard. The method involved isocratic elution on a Zorbax Eclipse XDB‐C 18 column and mass spectrometry in positive multiple reaction monitoring mode, achieving good linearity over 0.4–600 ng/mL. The method demonstrated intra- and inter-day precisions with relative standard deviations of less than 13.5% and accuracy between 94.2% and 97.5%. It was successfully applied to study the pharmacokinetics of AA‐I in rats.
