Keratinocytes maintain compartmentalization between dermal papilla and fibroblasts in 3D heterotypic tri‐cultures

    The study found that keratinocytes are essential in maintaining the separation between dermal papilla (DP) cells and human dermal fibroblasts (HDF) in a three-dimensional (3D) cell culture environment. Using a tri-culture model with DP cells, HaCaT keratinocytes, and HDF cells in polyethylene glycol diacrylate (PEGDA) hydrogel microwells, the researchers observed that keratinocytes formed a barrier around DP cells, preserving their aggregated state which is crucial for hair follicle development. The 3D cultures also showed increased expression of DP genes compared to two-dimensional cultures, indicating a more in vivo-like condition. The study highlights the role of keratinocytes in hair follicle formation by preventing the mixing of DP and HDF cells, which could be significant for future hair follicle research and regenerative medicine applications.
