TLDR Type I interferons play a key role in the development of various skin diseases.
The document reviewed the role of Type I interferons (IFNs), which are cytokines involved in immune responses against infections, in both healthy skin and various autoimmune and inflammatory skin diseases. It highlighted that Type I IFNs, including IFNα, IFNβ, and IFNκ produced by the epidermis, are significant in the pathogenesis of diseases such as cutaneous lupus erythematosus (CLE), dermatomyositis, Sjogren’s syndrome, psoriasis, lichen planus, alopecia areata, and vitiligo. The review, based on a PubMed database search, discussed the impact of IFNs on keratinocyte biology and the genetic and protein changes induced by IFNs that contribute to clinical manifestations of these skin conditions.

FCE 28260 (PNU 156765), an under-explored 5α-reductase inhibitor, showcases promising results in research by Giudici et al., outperforming well-known treatments like Finasteride in reducing the conversion of testosterone to DHT. Its superior efficacy, demonstrated through lower IC50 values in both natural and human recombinant enzyme studies, suggests it could offer more effective management of DHT-related conditions. Additionally, its lower molecular weight hints at better potential for topical application, potentially offering advantages in treating conditions such as androgenic alopecia. Despite its potential, it has not advanced in development, possibly due to financial limitations, leaving its therapeutic prospects and side effect profile largely unexplored.

Excess sebum rich in cholesterol and triglycerides can lead to hair loss through inflammation, with treatments like Ciclopirox shampoo, Benzoyl Peroxide shampoo, and Clindamycin gel recommended for managing conditions like seborrheic dermatitis and folliculitis. Pioglitazone is suggested for Lichen Planopilaris, while Omega-3s and reducing processed foods may improve sebum quality, though genetic factors play a significant role.
