High-Voltage Electron Microscopy and 3D Reconstruction of Human Hair Fiber Ultrastructure

    The study investigated the ultrastructure of the non-keratinous regions of human hair fibers, specifically the endocuticle, the cell membrane complex, and the nuclear remnants of the cortex, using conventional and high voltage electron microscopy. The aim was to understand how external factors affect hair by examining these regions in detail. To avoid artifacts caused by thin sectioning, a three-dimensional reconstruction was performed using serial, semi-thick sections. Hair fibers were collected from a female subject and were untreated by chemicals. Samples were embedded, sectioned, stained, and examined using a high voltage electron microscope. The study provided a clearer characterization of the non-keratinous regions of human hair fibers.
